I'm not sure how to phrase this as a simple question, so I'm gonna use examples;
Say, I wanted to play as a Youtuber, for this example I'mma use, Markiplier. That's an obvious "no,"
But, say, if I wanted to play as a version of Markiplier, from one of his fangames, for instance, Dream.Exe. There is nothing written for or against this, at least that I read.
Would that be okay?
((As a note, yes, that is legitimately an idea of mine for my new character.))

So you're asking if you can play the 'character/persona' that person plays in a specific piece of media rather than them themselves?

Not...exactly, but that's somewhat close.
Dream.exe is a Markiplier fan-game made in RPG Maker. You play as Markiplier himself, but Markiplier had no involvement in development besides inspiration.
My question is, can you play as that in-game 'Markiplier,' as long as you aren't playing as real-life Markiplier?

I am inclined to say no. At least based on your description, Markiplier from Dream.exe is essentially just the real Markiplier placed into a video game. A real-life figure appearing in a piece of media as themselves is not justification for being able to use that person as a character, since practically all aspects of the 'character' are essentially just the qualities and characteristics of the real-life figure.
As a general rule, the more the 'character' is an entity onto itself, separate from its creator, the more likely we are to permit it as a character in the Omniverse. The only potential exception to this rule is if a member (somehow) has explicit permission from the person in question to roleplay as them.
