Costa and the surrounding waters are pretty Coruscant-heavy, with stormtroopers and the whatnot. Given the size and contents of the verse and the city being at the gate, I imagine they would want to heavily secure the tower.
Greg is the One Piece guy, so I can only assume that the prison would be similar to how it is in the canon/wiki. I'm sure there are probably differences, given this is the Omniverse (there'd probably be stormtroopers, space marines, and others from Coruscant along with locals from Vasty Deep). You could attempt a jailbreak, but remember that such an action, whether it ends in failure or success, would result in a bounty being placed on your head. Probably a hefty one given the status of the prison.
I think Mewtwo is supposed to be taken from right after the end of the movie. I never watched the show, so I dunno if he appears in that or the manga, but I imagine it'd be safe to guess it's from the end of the movie, since he's supposed to be the 'good' leader of a neutral faction.

In general, I think it depends on the roaming NPC in question whether you can just casually encounter them or require staff input. Some, like Baron Victor, will be simple to find since he's a public figure with a loose schedule, or mercenaries like Whompt. Some are harder, given their lifestyle.
